A selection layer is a static snapshot of a selection.  It will not change if the data are updated.  The user cannot add or remove features from the selection in the selection layer.  A definition query is based on an attribute query.  Every time the layer is redrawn, the query is run again, so it is a 'live' subset of data.  If the data are updated, features can be added or removed from the query automatically.
